T7启动子-11碱基突变对转录影响的研究

摘要:
T7噬菌体启动子能被T7RNA聚合酶和真核生物RNA聚合酶Ⅱ系统启动转录,为研究两个系统转录的关键碱基,将合成的T7噬菌体启动子-11变异体与报道基因CAT基因连在一起。体内CAT和体外狭缝RNA杂交实验显示:-11碱基是T7RNA聚合酶和真核生物RNA聚合酶Ⅱ系统启动T7启动子的关键碱基之一。

The Effects of Mutant T7 Promoter on Transcriptions
Abstract:
To determine key bases on T7 promoter recognized by T7 RNApolymerase and eukaryotic RNA polymerase, the base located at -11 of the promoter was replaced with other bases and the mutant promoters were inserted upstream of coding sequences of CAT gene. CAT test in vivo and Slot-Blot test in vitro showed that the promoter could not be transcripted by both T7 RNA polymerase and eukaryotic RNA polymerase Ⅱ.The result suggests that the -11 base might be the key base for T7 RNA polymerase and eukaryotic RNA polymerase Ⅱsystems.
