向日葵花盘水提物对硫色镰刀菌及其细胞壁降解酶活性抑制特性

摘要:
通过抑菌及细胞壁降解酶活性试验,研究向日葵花盘(sunflower disc,SFD)水提物对引起马铃薯干腐病主要病原菌——硫色镰刀菌(Fusarium sulphureum)生长及其侵染不同马铃薯品种时分泌的多聚半乳糖醛酸酶(polygalacturonase,PG)、果胶甲基半乳糖醛酸酶(polymethyl-galacturonase,PMG)、纤维素酶(carboxymethyl cellulose,Cx)、β-葡萄糖苷酶活性变化。结果表明,向日葵花盘水提物对硫色镰刀菌生长及侵染陇薯7号、陇薯10号、新大坪3个马铃薯品种分泌的4种细胞壁降解酶活性均具有抑制作用,最佳抑制浓度为320 mg/mL,在0~640 mg/mL浓度内,随浓度增大,对硫色镰刀菌生长的抑制作用增强;经最佳抑制浓度处理能使陇薯7号的PG和β-葡萄糖苷酶活性降低到未侵染水平,但β-葡萄糖苷酶活性的降低未达到极显著效果,该处理不能使陇薯7号的PMG和Cx及陇薯10号、新大坪的4种细胞壁降解酶活性降低到未侵染水平。

Abstract:
By methods of bacteriostatic test and cell-wall degrading enzyme activity test, this study investigated the biological control effect of water extract from sunflower disc on Fusarium sulphureum and the activities of 4 cell-wall degrading enzymes. Polygalacturonase(PG), polymethyl-galacturonase(PMG), carboxymethyl cellulose(Cx) and β-glucosidase were 4 cell-wall degrading enzymes secreted by Fusarium sulphureum invading the different potato varieties. Results show that the extract is of inhibition effects on Fusarium sulphureum and activities of 4 cell-wall degrading enzymes secreted by Fusarium sulphureum invading potato tubers from 3 potato varieties of Longshu No. 7, Longshu No. 10 and Xindaping, and the best inhibition occurrs at the concentration of 320 mg/mL. With the increasing of concentration in the range of 0 to 640 mg/mL, the inhibition effect on Fusarium sulphureum increased. Treated by the extract of the best concentration, the activities of polygalacturonase(PG) and β-glucosidase of Longshu No. 7 can be reduced to the uninfected level, but the reduction of β-glucosidase activity did not achieve a very significant effect. The activities of polymethyl-galacturonase(PMG) and carboxymethyl cellulose(Cx) of Longshu No. 7 and the activities of 4 cellwall degrading enzymes of Longshu No. 10 and Xindaping could not be reduced to uninfected levels.
