采用蔗糖密度梯度离心法(95000×g,2 h)提取大鼠肝细胞过氧化物酶体,所得过氧化物酶体形态完整,纯度与肝匀浆相比提高了26倍,仅有少量(0.5%~0.9%)的微粒体和线粒体污染,回收率为12%.为研究过氧化物酶体提供了有效的分离方法.此法还可将过氧化物酶体、微粒体、线粒体同时进行分离. |

The Isolation of Peroxisomes from rat liver |

Abstract: |
A modified method for isolating of peroxisomes from rat liver was described. The peroxisome enriched fraction was put onto discontinuous sucrose density gradients and then centrifuged for 2 h at 95000×g. It was found that the peroxisomes obtained with this method were intact, and contained only minute amounts (0.5%~0.9%) cellular mitochondria and microsomes. The purity of the peroxisome was increased 26 fold compared with the liver homogenate. The final yield of peroxisomes was about 12%. The peroxisomes, mitochondria and microsomes could be separated at the same time by this method which provided a basis for the study of peroxisomes. |
