基于线粒体Cyt b和D-loop区的雅鲁藏布江中上游黑斑原鮡遗传多样性分析

摘要:
为了解西藏雅鲁藏布江中上游黑斑原鮡(Glyptosternon maculatum)群体的遗传多样性,为黑斑原鮡的保护和人工选育提供理论依据。本研究以雅鲁藏布江中上游黑斑原鮡群体为研究对象,通过对其线粒体细胞色素b(Cytochrome b,Cyt b)和控制区(D-loop区)基因序列进行扩增和测序,分析了黑斑原鮡的线粒体序列特征与其群体遗传多样性的关联。结果显示,黑斑原鮡Cyt b基因序列长度为1138 bp,A+T含量(59.5%)高于C+G含量(40.5%),检测到变异位点32个,其中单变异位点26个,简约信息位点6个,共检测到21个单倍型,其中单倍型Hap1个体数最多,占总检测个体数的69.44%,群体单倍型多样性(Hd)为0.509±0.050,核苷酸多样性(Pi)为0.00071±0.00011,Tajima's D中性检验值为-2.53291,中性检验结果显示(P<0.001)极显著负值;D-loop区基因序列长度为893 bp,A+T含量(64.31%)高于C+G含量(35.69%)共检测到变异位点35个,包括单变异位点21个,简约信息位点14个,检测到23个单倍型,Hd为0.701±0.028,Pi为0.00167±0.00028,单倍型Hap 1和Hap 3的个体数较多,分别占45.14%和30.56%,D-loop基因Tajima's D中性检验值为-2.36638,中性检验结果显示(P <0.01)极显著负值,表明该群体经历过种群扩张。黑斑原鮡群体的Cyt b和D-loop区基因适合进行黑斑原鮡的种内遗传分析,且D-loop区基因更具有进化优势,其群体的遗传多样性较低,为黑斑原鮡遗传育种与保护提供一定的参考价值。

Genetic diversity analysis of Glyptosternon maculatum in the upper and middle reaches of the Yarlung Zangbo River based on mitochondrial Cyt b and D-loop regions

Abstract:
To comprehend the genetic diversity of Glyptosternon maculatum in the upper and middle reaches of the Yarlung Zangbo River in Tibet, and to establish a theoretical foundation for the conservation and artificial breeding of Glyptosternon maculatum. In this study, Cytochrome b (Cyt b) and D-loop gene sequences were amplified and sequenced from specimens collected in the upper and middle reaches of the Yarlung Zangbo River. Subsequently, the correlation between mitochondrial sequence characteristics and the genetic diversity of the population was analyzed. The findings revealed that the gene sequence length was 1138 bp, with a higher A+T content (59.5%) compared to C+G (40.5%). Thirty-two mutation loci were identified, comprising 26 single mutation loci and 6 psionic information loci. A total of 21 haplotypes were identified, with Hap1 being the most prevalent. Population haplotype diversity (Hd) was measured at 0.509±0.050, nucleotide diversity (Pi) at 0.00071±0.00011, and Tajima's D neutral test value at -2.53291 (P<0.001).The D-loop sequence was 893 bp long, with a higher A+T content (64.31%) than C+G (35.69%). Thirty-five mutation sites were detected, including 21 single mutation sites and 14 reduced information sites, resulting in 23 haplotypes. Hd was calculated at 0.701±0.028, Pi at 0.00167±0.00028. Haplotypes Hap1 and Hap3 were more prevalent, constituting 45.14% and 30.56% of individuals, respectively. Tajima's D neutral test value for the D-loop gene was -2.36638, indicating extremely significant negative values (P<0.01) and suggesting population expansion in this population.The Cyt b gene and D-loop region within the population proved suitable for intraspecific genetic analysis Particularly, the D-loop region gene exhibited greater evolutionary advantages despite the population's lower genetic diversity.These findings offer valuable insights for genetic breeding and the conservation of Pareuglis melanoglanis.
