胡杨根际土壤来源放线菌分离及其拮抗活性筛选

摘要:
根际土壤微生物在抵御病原微生物的入侵、提高植物的抗病能力等方面具有重要作用。胡杨作为沙漠地区的常见植物,其根际土壤微生物种类丰富且具有很高的研究价值。为了探究根际土壤微生物的作用,丰富和完善极端环境拮抗活性微生物菌种库,本研究基于选择培养基筛选法分析了新疆南疆地区10种不同地区的胡杨根际土壤的放线菌资源多样性,并采用细菌的16S rDNA测序和平板对峙法筛选。结果表明:经过分离与筛选,共获得269株菌株,选择生长状态较好的78株进行分析鉴定,分别为5个属,其中链霉菌属(Streptomyces)为优势类群。利用平板对峙法和琼脂扩散法以白色念珠菌(Candida albicans)、细极链格孢菌(Alternaria tenuissima)、大丽轮枝菌 Verticillium dahliae)、尖孢镰刀菌(Fusarium oxysporum)、金黄色葡萄球菌(Staphylococcus aureus)、铜绿假单胞菌(Pseudomonas aeruginosa)、大肠杆菌(Escherichia coli)、肺炎克雷伯菌 Klebsiella pneumoniae)、解淀粉欧文氏菌(Erwinia amylohydrolylic)、伤寒沙门氏菌(Salmonella typhi)为靶标菌,对分离出来的菌株进行拮抗活性检测,筛选出32株活性放线菌。这些结果丰富了微生物资源并为新疆主要农牧业病害的防控提供了菌种基础。

Isolation of actinomycetes from rhizosphere soil of Populus euphratica and screening of their antagonistic activity

Abstract:
Rhizosphere soil microorganisms are crucial in resisting the invasion of pathogenic microorganisms and enhancing plant disease resistance. Populus euphratica, a commonly found plant in desert areas, possesses a diverse range of rhizosphere soil microorganisms and holds significant research value. This study aims to investigate the microbial interactions in the rhizosphere soil of Populus euphratica and enrich the extreme environmental antagonistic microbial strain library. The diversity of actinomycete resources in 10 different regions of Populus euphratica rhizosphere soil in southern Xinjiang was analyzed using the selective medium screening method. The strains were subjected to 16S rDNA sequencing and plate confrontation method for screening. The results revealed the isolation and screening of a total of 269 strains, with 78 strains exhibiting good growth status selected for analysis and identification. These strains were classified into 5 genera, with the genus Streptomyces being the dominant group. Furthermore, 32 strains of active actinomycetes were screened for antagonistic activity, and targeting pathogens included Candida albicans, Alternaria tenuissima, Verticillium dahliae, Fusarium oxysporum, Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae, Erwinia amylohydrolylic, and Salmonella typhi. These findings contribute to the enrichment of microbial resources and provide a microbial strain foundation for the prevention and control of major agricultural and animal husbandry diseases in Xinjiang.
