骆驼刺内生真菌的分离、鉴定与抑菌活性初筛

摘要:
本研究采集干旱沙漠植物骆驼刺(Alhagi sparsifolia Shap.)标本,采用匀浆涂布法分离培养骆驼刺内生真菌,共分离获得9株内生真菌,通过ITS序列测定的方法进行鉴定,其中7株(LTCZ2、LTCY1a、LTCY1b、P9、P10、P11、3P1)为外担菌纲(Exobasidiomycetes),2株(LTCY2、LTCY3)为不整囊菌纲(Eurotiomycetes)曲霉科(Eurotiaceae)曲霉菌属(Aspergillus)。采用菌饼法检测其抑菌活性,P9、P10、P11、LTCY1a、LTCY1b、LTCZ2、LTCY3对金黄色葡萄球菌(Staphylococcus aureus)有潜在抑菌活性,LTCY3、P10和LTCY2对粪肠球菌(Enterococcus faecalis)有潜在抑菌活性,其中分离获得的一株曲霉属内生真菌LTCY3对临床常见的致病菌金黄色葡萄球菌、大肠埃希菌(Escherichia coli)、铜绿假单胞菌(Pseudomonas aeruginosa)、粪肠球菌均具有潜在抑菌活性。本研究以期为今后开发利用新的微生物资源提供科学依据。

Isolation, identification and primary screening of antibacterial activity of endophytic fungi of Alhagi sparsifolia Shap.

Abstract:
In this study, the samples were collected from arid desert plant Alhagi sparsifolia Shap., and its endophytic fungi were isolated and cultured by uniform coating method. 9 strains of endophytic fungi were isolated and identified by ITS sequencing, of which 7 strains(LTCZ2, LTCY1a, LTCY1b, P9, P10, P11, 3P1) belonged to Exobasidiomycetes and 2 strains(LTCY2, LTCY3) belonged to Aspergillus sp. The colony cake method was used to detect the antibacterial activity. P9, P10, P11, LTCY1a, LTCY1b, LTCZ2 and LTCY3 have potential antibacterial activity against Staphylococcus aureus, and LTCY3, P10 and LTCY2 have potential antibacterial activity against Enterococcus faecalis. We found an endophytic fungus LTCY3 (Aspergillus sp.) has obvious antibacterial activity against common clinical pathogens Staphylococcus aureusEscherichia coliPseudomonas aeruginosa, and Enterococcus faecalis. This study is expected to provide scientific basis for future development and utilization of new microbial resources.
