僵蚕药材酸溶性蛋白质的分离提取及鉴定

摘要:
以Box-Behnken组合设计进行僵蚕药材酸法提取工艺的响应面优化,并对提取物中蛋白质进行定性定量分析。结果表明:酸法工艺获取僵蚕蛋白质的最佳条件为提取温度31.8 ℃、提取时间3.0 h、液料比49.6∶1(Vm)、醋酸钠缓冲液浓度0.12 mol/L,此时蛋白质提取率为5.34%。十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)指纹图谱结果表明,僵蚕酸溶性蛋白质主要分布于26 390~31 570和59 130~81 790区间,其中31 570与27 870蛋白约占总蛋白的59.9%,71 700与81 790蛋白约占总蛋白的24.3%。傅里叶变换红外吸收光谱(FTIR)结果表明,僵蚕酸溶性蛋白质具有特征性指纹图谱。上述结果有望用做僵蚕药材分子鉴定的依据,有助于僵蚕资源的开发利用。

Isolation and identification of acid-soluble proteins of Bombyx Batryticatus

Abstract:
The response surface method of Box-Behnken design was used to optimize the acid extraction process of proteins from Bombyx Batryticatus extract, and the corresponding protein fingerprint was established. The results showed that the best extraction conditions were as follows: the extraction temperature at 31.8 ℃, the extraction time 3.0 h, the liquid-to-material ratio at 49.6∶1(Vm), and the sodium acetate buffer concentration 0.12 M. Under these conditions, the protein extraction rate was 5.34%. SDS-PAGE fingerprint results showed that the acid-soluble proteins of the Bombyx Batryticatus were mainly distributed in the 26 390~31 570 and 59 130~81 790 intervals, of which 31 570 and 27 870 accounted for about 59.9% of the total proteins, and 71 700 and 81 790 proteins accounted for about 24.3%. The results of FTIR showed that the acid-soluble proteins of the Bombyx Batryticatus had a characteristic fingerprint. The above results are expected to be used as the basis for the development of the resources and the molecular identification of Bombyx Batryticatus medicinal materials.
