脱氢弯孢霉菌素对脂多糖诱导的巨噬细胞炎症研究

摘要:
研究脱氢弯孢酶菌素(Dehydrocurvularin, DCV)对脂多糖(lipopolysaccharide,LPS)诱导的小鼠腹腔(peritoneal macrophages, PMs)和骨髓来源的巨噬细胞(bone-marrow-derived macrophages, BMDMs)的影响。MTT方法筛选最佳药物浓度处理不同来源的巨噬细胞,采用Western blot方法检测诱导型一氧化氮合酶(induceble nitric oxide synthase, iNOS)、环氧化酶-2(cyclooxygenase-2, COX-2)及炎性信号通路NF-κB和MAPK的相关蛋白的表达。结果显示,脱氢弯孢酶菌素可显著抑制LPS诱导的iNOS、COX-2的表达,且呈剂量依赖性。NF-κB和MAPK信号通路相关的蛋白激活也受到了明显的抑制。结果表明,脱氢弯孢酶菌素可显著抑制LPS诱导的不同来源的免疫细胞的炎症。

LPS-simulated inflammatory response by dehydrocurvularin in macrophages

Abstract:
To study the effect of dehydrocurvularin (DCV) in lipopolysaccharide (LPS)-induced mouse peritoneal macrophages (PMs) and bone-derived macrophages (bone-marrow-derived macrophages, BMDMs). MTT was used to screen the optimal drug concentration to treat macrophages from different sources. Western-blot method was used to detect cyclooxygenase-2 (COX-2) and inflammatory signaling pathway NF-κB and MAPK related proteins. DCV significantly inhibited LPS-induced COX-2 expression in a dose-dependent manner. Protein activation associated with NF-κB and MAPK signaling pathways was also significantly inhibited. DCV inhibits LPS-induced inflammatory properties of immune cells from different sources.
