野生大黑蚁中抗菌肽类物质的分离纯化及其耐热性 |

摘要: |
以野生大黑蚁为原料,采用索氏抽提法除脂后,经乙醇浸提法粗提,再利用透析法分离纯化,得到对细菌具有抑制作用的抗菌肽类物质,并对该抗菌肽类物质进行氨基酸分析和耐热性检测。结果表明:分离得到的抗菌肽类物质对枯草芽胞杆菌具有抑制作用,其抑菌圈直径为24 mm;结合索氏抽提除脂,乙醇浸提和透析法所得到的粗提物氨基酸含量较高;蚂蚁抗菌肽类物质表现出较高的热稳定性,80 ℃下加热50 min后抑菌活性下降。 |

The wild black ant Polyrhachis vicina Roger was used as raw material, which was degreased by Soxhlet extraction. After crude extraction by ethanol, it was separated and purified by dialysis method to obtain antimicrobial peptides with antibacterial effect. Amino acid analysis and heat resistance detection of the antimicrobial peptide substances were performed.The results showed that the isolated antibacterial peptide had an inhibitory effect on Bacillus subtilis, and the diameter of its bacteriostasis was 24 mm. The ant antibacterial peptide obtained by Soxhlet extraction combined with ethanol extraction and dialysis had higher amino acid content, and showed higher thermal stability, the antibacterial activity decreased after heating at 80 ℃ for 50 min and more. |
