小花清风藤的化学成分及其对肝细胞的保护作用

摘要:
采用正相硅胶柱色谱、Sephadex LH-20柱色谱、重结晶等多种分离/纯化方法,利用核磁共振(NMR)波谱分析法,对小花清风藤茎叶的化学成分进行分离与鉴定。结果从小花清风藤茎叶的石油醚萃取部位分离并鉴定出4个化合物,包括2个五环三萜类化合物和2个其他类化合物,分别为24-羟基-3-氧代-12-齐墩果烯-28-酸(化合物1),β-谷甾醇(化合物2),豆甾醇(化合物3)和齐墩果酸(化合物4)。其中,化合物1为首次从该属植物中分离得到。采用四唑盐(MTT)法测试石油醚萃取物和分离得到的4个化合物对CCl4损伤的LO2肝细胞的保护作用,结果均显示出良好的肝细胞保护作用,其中石油醚部位和化合物4肝细胞保护作用最明显,能够显著提高CCl4损伤LO2肝细胞的存活率。

Abstract:
Various means such as silica gel, Sephadex LH-20, and recrystallization were used for separation and purification of chemical constituents from the stems and leaves of Sabia parviflora, and their structures were identified by nuclear magnetic resonance (NMR) spectra data. Four compounds were obtained from the petroleum ether extract of Sabia parviflora, including two pentacyclic triterpenes and two other compounds, and their structures were identified as 24-hydroxy-3-oxoolean-12-en-28-oic acid (1), β-sitosterol (2), stigmasterol (3) and oleanolic acid (4), respectively. Compound 1 was isolated from Sabia parviflora for the first time. Methyl thagolyl tetragoliam (MTT) assay was used to determine the protective effects of petroleum ether extract and four compounds on hepatocyte injury induced by CCl4 in LO2 cells. The results showed that all samples had hepatocyte protection activities. Among them, the petroleum ether extract and compound 4 had better hepatocyte protection activities, and they could significantly improve the survival rate of LO2 hepatocytes injured by CCl4.
