多肉植物糯玉露离体再生影响因子研究

摘要:
本研究以糯玉露的叶片为起始外植体,对其进行了离体再生过程中相关影响因子的生物学研究,旨在建立糯玉露高效离体再生快速繁殖体系。研究结果表明:糯玉露诱导愈伤组织的最佳α-萘乙酸(NAA, 1-naphthylacetic acid)浓度为1.5 mg/L,出愈率达87.30%,愈伤组织比重为0.74 g/cm3;最适宜的不定芽诱导培养基为MS+1.0 mg/L 6-苄氨基腺嘌呤(6-BA,6-Benzylamino purine)+0.3 mg/L NAA,不定芽诱导率为93.19%,平均不定芽数为8.6个;最适宜的不定芽伸长培养基为MS+0.3 mg/L 6-BA+1.0 mg/L 赤霉素(GA3,gibberellin A3),不定芽伸长率为74.67%,芽的均伸长长度为2.26 cm;最适宜的生根培养基为1/2MS+2.0 mg/L NAA +0.5 mg/L 吲哚丁酸(IBA,3-indolebutyric acid),生根率可达92.18%,移栽存活率可达62.19%。本研究以糯玉露叶片为外植体,建立了离体再生体系,提高了多肉植物糯玉露的繁殖效率,以期满足多肉植物糯玉露的市场产业需求。

Study on influence factors of in vitro regeneration of Haworthia cooperi Baker ‘Nuoyulu’

Abstract:
In this study, the leaves of ‘Nuoyulu’ was used as the initial explants, and biological studies were carried out on related factors affecting in vitro regeneration to establish an efficient regeneration system for Haworthia cooperi Baker ‘Nuoyulu’. The results showed that the optimum 1-naphthylacetic acid (NAA) concentration in callus occurrence was 1.5 mg/L and the callus induction rate was 87.30%, with the callus proportion of 0.74 g/cm3. The optimum medium for the adventitious bud occurrence was MS+1.0mg/L 6-benzylamino purine (6-BA)+0.3 mg/L NAA. The frequency of differentiation was 93.19%, and average number of adventitious buds was 8.6. The suitable medium for adventitious bud elongation was MS+0.3 mg/L 6-BA+1.0 mg/L gibberellin A3 (GA3),the elongation efficiency was about 74.67%, and the average shoot length was 2.26 cm. The optimum rooting medium was 1/2MS+2.0 mg/L NAA+ 0.5 mg/L 3-indolebutyric acid (IBA),the rooting rate reached up to 92.18%, and the survival rate of transplanting reached up to 62.19% .The conclusion is that the regeneration system of the leaves of ‘Nuoyulu’ in vitro is established, which improves the reproductive efficiency of Haworthia cooperi Baker ‘Nuoyulu’ and should satisfy the demand of the market industry.
