巨桉PIF基因家族的鉴定及表达模式分析

摘要:
光敏色素作用因子(phytochrome interacting factors,PIF)是bHLH家族中的关键转录因子。目前,PIF基因已在拟南芥、玉米、油菜等植物中得到了广泛的研究,已证实其在植物生长发育、光反应及逆境响应过程中具有重要作用。然而,巨桉(Eucalyptus grandis)中PIF基因家族成员的系统鉴定与分析尚未报导。本研究对桃金娘科速生树种巨桉PIF家族基因的特征和进化进行了研究,从巨桉中鉴定出10个巨桉PIF基因(EgPIF1-EgPIF10),编码的氨基酸数量在405-660 aa之间,分子质量范围为44.6-70.0 kDa,等电点(PI)为5.41-8.73;染色体定位分析显示EgPIF不均匀地分布在4条染色体上。多序列比对结果表示,EgPIF均存在bHLH保守结构域;启动子区域分析表明EgPIF基因含有光响应元件。分析了EgPIF基因在茉莉酸甲酯(MeJa)、氯化钠(NaCl)和水杨酸(SA)处理下的表达模式,结果显示,EgPIF1-3/5在不同的处理下一直表现出较高的表达水平,表明其在应对各种逆境中发挥重要作用。上述研究结果可为进一步探究EgPIF基因家族成员在巨桉的生长发育调控和信号转导通路中的功能奠定基础。

Identification and expression pattern analysis of the PIF gene family in Eucalyptus grandis

Abstract:
Phytochrome-interacting factors (PIFs) are key transcription factors belonging to the bHLH family. Currently, PIF genes have been extensively studied in plants such as Arabidopsis thaliana, maize(Zea mays L.), and rapeseed(Brassica napusL.), and their crucial roles in plant growth and development, photoresponse, and stress adaptation have been well established. However, systematic identification and analysis of PIF gene family members in Eucalyptus grandis have not yet been reported. This study investigated the characteristics and evolution of the PIF gene family in E. grandis, a fast-growing species of Myrtaceae family. A total of 10 E. grandis PIF genes (EgPIF1-EgPIF10) were identified, encoding proteins ranging from 405 to 660 amino acids in length, with molecular weights between 44.6 and 70.0 kDa and isoelectric points (pI) ranging from 5.41 to 8.73. Chromosomal localization analysis revealed that EgPIFs are unevenly distributed across four chromosomes. Multiple sequence alignment indicated that all EgPIFs contain the conserved bHLH domain. Promoter region analysis demonstrated that EgPIF genes contain light-responsive cis-elements. Expression patterns of EgPIFs under methyl jasmonate (MeJA), sodium chloride (NaCl), and salicylic acid (SA) treatments were analyzed. The results showed that EgPIF1, EgPIF2, EgPIF3, and EgPIF5 consistently exhibited relatively high expression levels under different treatments, suggesting their important roles in responding to various stresses. These findings lay a foundation for further investigation into the functions of EgPIF gene family members in regulating growth and development and signal transduction pathways in E. grandis.
