大黄内生真菌产大黄素类物质的研究

从西宁市二十里铺村植物园中采集了一株野生大黄,将大黄的根、茎、叶表面消毒后,共分离出72株内生真菌。从大黄内生真菌中筛选出一株产大黄素类物质的内生真菌,从大黄根茎叶中筛选出的大黄内生真菌进行发酵培养,用氯仿萃取发酵液中的大黄素,再用薄层层析法定性分析,高效液相色谱定量分析。从大黄根的内生真菌中分离筛选到一株产大黄素含量最高的内生真菌GP15。对该菌株的代谢产物研究表明,其代谢产物中含有大黄素的量为18mg/L,充分说明利用大黄内生真菌发酵产大黄素是可行的。

Study on productin of emodin by endophytic fungi from Rhubarb

Abstract:
:In this study, we collected a wild Rhubarb from the Botanic Garden in Xining Ershilipu village. After surface disinfected of Rhubarb root,stem and leaves,72 endophytic fungi strains were isolated. An endophytic fungus producing emodin was selected from Rhubarb endophyte,endophytic fungi screened from Rhubarb roots、stems、and leaves were fermented and cultured.Extraction of emodin from fermentation broth with chloroform,through thin layer chromatorgraphy and high performance liquid chromatography to select which Rhubarb endophyte fungi strain can produce emodin .Then thin layer chromatorgraphy was used for qualitative analysis and high performance liquid chromatography for quantitative analysis. Isolation and screention endophytic fungi GP15 from the Rhubarb root,which can produce the highest emodin . The strain GP15 fermentation broth showed that its metabolites containing emodin 18mg/L , fully demonstrated that using endophytic fungi fermentation broth to produce emodin is possible. .
